When is a good time to visit? Will august school holidays be good?

School holidays are the best if you want a discounted rate (esp end of the year long holidays). I went to Singapore zoo about a year ago and only paid about half of the usual admission rate- have to show passport to proof Malaysian. Got a little embarrassed as their Singaporean had to pay the full price.
Expect a full day adventure and therefore would be better to put up a night/s.
Can fly air asia now..

If driving, then to use the second link (Journey from KL to Tuas would take about 4 hours +).
And you would want to buy(optional for convenience) a 'smart tag' with a credit sort of thing with a scanner (to be installed in car) to autodebit when you pass a gantry or entering car parks...

Is the promotion open for every Malaysian school holidays? I heard about it last year or so whereby we pay RM18 instead of SGD 18...

They have been having this sort of promotion annually for quite some time now. And you are right- you can pay in RM but I am not so sure if it is SGD18 to RM18. Maybe it is like RM25 or SGD 10 or something like that.
Can't remember.
